  1. Tóm tắt nội dung luận án

Kiên Giang có hệ sinh thái đa dạng với nhiều loài thực vật được sử dụng làm thuốc. Trong nghiên cứu này, 23 loài thực vật thu hái tại một số huyện của tỉnh Kiên Giang được ngâm chiết với dung môi ethanol 96. Các cao chiết ethanol được tiến hành đánh giá hoạt tính kháng oxi hóa in vitro để chọn lựa loài thực vật có hoạt tính mạnh. Kết quả sàng lọc cho thấy cây Lý (Syzygium jambos), Luân thùy cambot (Spirolobium cambodianum) và Chùm đuông (Sphaerocoryne affinis) có hoạt tính kháng oxi hóa mạnh ở bốn phương pháp thử nghiệm là DPPH, ABTS●+, RP và TAC với giá trị EC50 trong khoảng 4,89 ‒ 46,33 µg/mL. Tương tự, ba loài thực vật này cũng thể hiện hoạt tính kháng khuẩn mạnh với giá trị MIC trong khoảng 80 ‒ 640 µg/mL và hàm lượng tổng polyphenol 570,36 ‒ 857,24 mg GAE/g cao chiết, hàm lượng flavonoid 413,18 ‒ 798,71 mg QE/g cao chiết. Vì vậy, chúng được chọn để tiếp tục nghiên cứu thêm thành phần hóa học và hoạt tính sinh học.

Từ lá Lý (S. jambos) đã phân lập được 10 hợp chất tinh khiết gồm stigmasterol (SJ1), 5((8'Z,11'Z,14'Z)-heptadeca-8',11',14'-trien-1-yl) benzene-1,3-diol (SJ2), β-sitosterol-3-O-β-D-glucopyranoside (SJ3), quercetin (SJ4), 2-phenyl-4H-chromen-4-one (SJ5), myricetin (SJ6), gallic acid (SJ7), caffeic acid (SJ8), chavicol β-D-glucopyranoside (SJ9), rutin (SJ10). Trong đó ba hợp chất gồm SJ2 , SJ5, SJ9 lần đầu tiên phân lập từ loài thực vật này. Các cao chiết và một số hợp chất như gallic acid, caffeic acid, quercetin, myricetin từ lá cây Lý (S. jambos) được đánh giá hoạt tính chống stress oxi hóa trên tế bào đại thực bào RAW264.7. Kết quả cho thấy cao chiết ethyl acetate và các hợp chất có khả năng tăng cường sự biểu hiện của gen kháng oxi hóa, thông qua việc thúc đẩy con đường truyền tín hiệu Nrf2/HO-1. Ngoài ra, hợp chất myricetin, quercetin cũng thể hiện hoạt tính kháng oxi hóa mạnh ở phương pháp DPPH, ABTS●+ với giá trị EC50 trong khoảng 2,67 ‒ 30,53 µg/mL.

Từ lá Luân thùy cambot (S. cambodianum) đã phân lập được 10 hợp chất tinh khiết gồm 27-p-Z-coumaroyloxyursolic acid (SC1), lupeol (SC2), p-hydroxybenzoic acid (SC3), ursolic acid (SC4), tectoquinone (SC5), daucosterol (SC6), asiatic acid (SC7), 1-O-β-D-glucopyranosyl-(2S,3S,4R,9Z)-2-[(2'R)-2'-hydroxytetracosanoylamino]-octadec-9-en-1,3,4-triol (SC9), naringenin (SC8), quercetin (SC10). Đặc biệt, các hợp chất này lần đầu tiên được phân lập trong chi Spirolobium. Ngoài ra, cao chiết ethanol từ lá cây Luân thùy cambot thể hiện hoạt tính chống stress oxi hóa in vivo trên mô hình ruồi giấm vì thức ăn bổ sung cao chiết làm tăng tuổi thọ của ruồi giấm. Bên cạnh đó, các hợp chất SC2, SC4, SC7 kháng khuẩn Streptococcus agalactiae, Aeromonas hydrophila, Aeromonas dhakensis, Edwardsiella ictaluri với giá trị MIC là 25 ‒ 1500 µg/mL.

Từ trái Chùm đuông (S. affinis) đã phân lập được 6 hợp chất tinh khiết gồm pinocembrin (SA1), naringenin (SA2), p-coumaric acid (SA3), caffeic acid (SA4), 5-(E)-caffeoylquinic acid (SA5), 5-O-p-coumaroylquinic acid methyl este (SA6). Tương tự, từ lá Chùm đuông (S. affinis) đã phân lập được 7 hợp chất tinh khiết gồm allantoin (SAL1), (+)-catechin (SAL2), apigenin (SAL3), rutin (SAL4), isatin (SAL5), (S)-3-hydroxy-3-(2-oxopropyl)indolin-2-one (SAL6),  nicotiflorin (SAL7). Trong đó, 11 hợp chất này lần đầu tiên được phân lập trong chi SphaerocoryneSA2, SA3, SA4, SA5, SA6, SAL1, SAL3, SAL4, SAL5, SAL6, SAL7. Bên cạnh đó, cao chiết ethanol từ lá và trái Chùm đuông được đánh giá hoạt tính stress oxi hóa in vivo (trong điều kiện stress oxi hóa do hydrogen peroxide 10% và paraquat 20 mM). Kết quả cho thấy tuổi thọ của ruồi giấm nuôi trong môi trường bổ sung cao chiết được kéo dài hơn so với môi trường đối chứng. Các kết quả nghiên cứu này góp phần cung cấp thêm cơ sở khoa học cho các nghiên cứu sâu hơn để tạo ra các sản phẩm ứng dụng trong lĩnh vực hóa dược. Từ đó cung cấp cơ sở khoa học định hướng cho việc khai thác sử dụng và bảo tồn nguồn tài nguyên thực vật một cách hợp lý.

  1. Những kết quả mới của luận án

Lần đầu tiên ở Việt Nam, lá cây Lý (S. jambos) được nghiên cứu về thành phần hóa học và hoạt tính sinh học. Kết quả cho thấy ngoài các thành phần hóa thực vật đặc trưng cho chi và loài, loài S. jambos ở Kiên Giang cũng xuất hiện 3 hợp chất mới trong loài. Đồng thời, lần đầu tiên thử nghiệm hoạt tính kháng khuẩn gây bệnh trên động vật thủy sản và hoạt tính chống stress oxi hóa ở mức tế bào RAW264.7 của cao chiết. Trong đó, các cao chiết và hợp chất phân lập từ cây Lý có khả năng tăng cường sự biểu hiện của gen kháng oxi hóa, thông qua việc thúc đẩy con đường truyền tín hiệu Nrf2/HO-1.

Lần đầu tiên ở Việt Nam và trên thế giới thành phần hóa học và hoạt tính sinh học của Luân thùy cambot (S. cambodianum) được nghiên cứu. Từ lá của cây Luân thùy cambot đã phân lập và xác định cấu trúc của 10 hợp chất tinh khiết, lần đầu tiên được phân lập trong chi Spirolobium. Cao chiết và các hợp chất phân lập từ Luân thùy cambot có hoạt tính kháng khuẩn gây bệnh trên động vật thủy sản, kháng oxi hóa in vitro và chống stress oxi hóa in vivo trên mô hình ruồi giấm.

Lần đầu tiên ở Việt Nam thành phần hóa học và hoạt tính sinh học như kháng oxi hóa in vitro, chống stress oxi hóa in vivo trên mô hình ruồi giấm của cây Chùm đuông (S. affinis) được nghiên cứu. Đã phân lập và xác định cấu trúc của 13 hợp chất, trong đó có 11 hợp chất tinh khiết lần đầu được phân lập từ chi Sphaerocoryne.

  1. Các ứng dụng/khả năng ứng dụng trong thực tiễn, các vấn đề cần tiếp tục nghiên cứu

Luận án cung cấp thông tin khoa học về thành phần hóa học và hoạt tính kháng oxi hóa, kháng khuẩn của 23 loài thực vật tại tỉnh Kiên Giang. Kết quả này góp phần vào việc tìm kiếm các hợp chất tự nhiên có khả năng chữa bệnh từ các loài thực vật sẵn có ở địa phương. Kết quả nghiên cứu đã đóng góp thông tin về thành phần hóa học của 03 loài S. jambos, S. cambodianum, S. affinis làm giàu thêm kho tàng hợp chất thiên nhiên của thế giới nói chung và Việt Nam nói riêng.

  1. Content of thesis summary

Kien Giang province, Vietnam, has a diverse ecosystem with many plant species used as medicine. In this study, 23 plant species collected in this province were extracted by using ethanol 96. The extracts were evaluated for in vitro antioxidant activity to select plant species with strong antioxidant activity. The results showed that the extract of Syzygium jambos, Spirolobium cambodianum and Sphaerocoryne affinis had strong antioxidant activities in four assays (DPPH, ABTS●+, RP and TAC) with EC50 values of 4.89 ‒ 46.33 µg/mL. Similarly, these extracts also showed strong antibacterial activity (Streptococcus agalactiae, Aeromonas hydrophila, Aeromonas dhakensis, Edwardsiella ictaluri) with MIC values in the range of 80 ‒ 640 µg/mL. Total polyphenol and flavonoid content of these extracts were 570.36 ‒ 857.24 mg GAE/g extract and 413.18 ‒ 798.71 mg QE/g extract, respectively. Therefore, they were selected for further research on their chemical composition and biological activities.

Ten compounds have been isolated from S. jambos leaves including stigmasterol (SJ1), 5((8'Z,11'Z,14'Z)-heptadeca-8',11',14'-trien-1-yl) benzene-1,3-diol (SJ2), β-sitosterol-3-O-β-D-glucopyranoside (SJ3), quercetin (SJ4), 2-phenyl-4H-chromen-4-one (SJ5), myricetin (SJ6), gallic acid (SJ7), caffeic acid (SJ8), chavicol β-D-glucopyranoside (SJ9), rutin (SJ10). Three compounds, namely 5((8'Z,11'Z,14'Z)-heptadeca-8',11',14'-trien-1-yl) benzene-1,3-diol (SJ2), 2-phenyl-4H-chromen-4-one (SJ5), chavicol β-D-glucopyranoside (SJ9) were isolated for the first time from this plant species. Extracts (ethanol and ethyl acetate extract from leaves) and some compounds from S. jambos leaves, namely quercetin (SJ4), myricetin (SJ6), gallic acid (SJ7) and caffeic acid (SJ8), were evaluated for their anti-oxidative stress activity on RAW264.7 macrophage cells. The results showed that ethyl acetate extract and all compounds had the ability to enhance the expression of antioxidant genes, through promoting the Nrf2/HO-1 signaling pathway. In addition, myricetin and quercetin compounds also showed strong antioxidant activity in DPPH and ABTS●+ assays with EC50 values of 2.67 ‒ 30.53 µg/mL.

Ten compounds have been isolated from S. cambodianum leaves including p-Z-coumaroyloxyursolic acid (SC1), lupeol (SC2), p-hydroxybenzoic acid (SC3), ursolic acid (SC4), tectoquinone (SC5), daucosterol (SC6), asiatic acid (SC7), 1-O-β-D-glucopyranosyl-(2S,3S,4R,9Z)-2-[(2'R)-2'-hydroxytetracosanoylamino]-octadec-9-en-1,3,4-triol (SC9), naringenin (SC8), quercetin (SC10). Interestingly, all of these compounds were isolated for the first time in Spirolobium genus. Ethanol extract from S. cambodianum leaves showed anti-oxidative stress activity in vivo in a fruit fly model.  Lupeol (SC2), ursolic acid (SC4) and asiatic acid (SC7) were antibacterial against Streptococcus agalactiae, Aeromonas hydrophila, Aeromonas dhakensis, Edwardsiella ictaluri with MIC values of 25 ‒ 1500 µg/mL.

Six compounds have been isolated from S. affinis fruits including pinocembrin (SA1), naringenin (SA2), p-coumaric acid (SA3), caffeic acid (SA4), 5-(E)-caffeoylquinic acid (SA5), 5-O-p-coumaroylquinic acid methyl este (SA6). Seven compounds were isolated from S. affinis leaves including allantoin (SAL1), (+)-catechin (SAL2), apigenin (SAL3), rutin (SAL4), isatin (SAL5), (S)-3-hydroxy-3-(2-oxopropyl)indolin-2-one (SAL6),  nicotiflorin (SAL7). There were 11 compounds, namely SA2, SA3, SA4, SA5, SA6, SAL1, SAL3, SAL4, SAL5, SAL6 and SAL7, were isolated for the first time in Sphaerocoryne genus. Ethanol extracts from S. affinis leaves and fruits were evaluated for oxidative stress activity in vivo using fruit fly model. The results showed that the lifespan of fruit flies increased in the treatments supplemented with the extracts as compared the control one (without the extracts).

The results obtained from this study provide scientific information for further research and development of products to improve human health. In addition, it provides a scientific basis to guide the reasonable exploitation, use and conservation of natural plant resources.

  1. The novel aspects of the thesis

For the first time in Vietnam, leaves of S. jambos were studied for their chemical composition and biological activity. The results show that in addition to the phytochemical components specific to the genus and species, S. jambos in Kien Giang also has three new compounds in the species. At the same time, for the first time, the extract tested the antibacterial activity on aquatic animals and the antioxidant stress activity at the RAW264.7 cell. In particular, extracts and compounds isolated from S. jambos have the ability to enhance the expression of antioxidant genes by promoting the Nrf2/HO-1 signaling pathway.

For the first time in Vietnam and the World, the chemical composition and biological activities of  S. cambodianum are studied. From the leaves of S. cambodianum, 10 pure compounds were isolated and determined for the first time in the genus Spirolobium. Extracts and compounds isolated from S. cambodianum have antibacterial activity against pathogens in aquatic animals, anti-oxidation in vitro, and anti-oxidative stress in vivo in the Drosophila melanogaster Model.

For the first time in Vietnam, the chemical composition and biological activities such as anti-oxidation in vitro and antioxidant stress in vivo in the Drosophila melanogaster Model of  the Sphaerocoryne affinis were studied. Isolated and determined the structure of 13 compounds, including 11 pure compounds isolated for the first time from this genus.

  1. Application prospects and suggestions for further study

The thesis provides scientific information about the chemical composition and antioxidant and antibacterial activities of 23 plant species in Kien Giang province. The research result contributes to the search for natural compounds with medicinal properties from locally available plants. The research results have contributed information about the chemical composition of three S. jambos, S. cambodianum, S. affinis species, enriching the treasure trove of natural compounds of the world in general and Vietnam in particular.
