  1. Tóm tắt nội dung luận án

Nông dược hữu cơ thường có xu hướng lưu tồn lâu trong hệ sinh thái, trong sinh quyển và thường gây độc cho con người và môi trường. Diazinon đã cấm sử dụng đối với nhiều quốc gia trên thế giới. Tuy nhiên, nông dân vẫn sử dụng thuốc trừ sâu chứa hoạt chất Diazinon để phòng trừ dịch hại cây trồng. Nhiều nghiên cứu về phân hủy sinh học Diazinon trong đất ở các vùng ôn đới, nhưng chưa có nghiên cứu về giảm thiểu ô nhiễm Diazinon trong đất vùng nhiệt đới bằng con đường sinh học. Vì thế đề tài được thực hiện nhằm các mục tiêu :

(1) phân lập và khảo sát khả năng phân hủy hoạt chất diazinon của các chủng vi khuẩn bản địa phân lập từ các mẫu đất được thu từ ruộng canh tác chuyên màu và luân canh lúa-màu ở ĐBSCL.

 (2) đánh giá ảnh hưởng của các yếu tố môi trường như nhiệt độ, pH, nguồn carbon, nồng độ diazinon đến sự gia tăng mật số vi khuẩn và tốc độ phân hủy Diazinon của một số chủng vi khuẩn tuyển chọn.

(3) xác định ảnh hưởng của cơ cấu cây trồng lên khả năng phân hủy thuốc trừ sâu diazinon ở điều kiện thực tế ngoài đồng ruộng và trong điều kiện nhà lưới.

(4) xác định sự ảnh hưởng của cơ cấu cây trồng lên cấu trúc của cộng đồng vi khuẩn trong điều kiện nhà lưới.

Phân lập vi khuẩn có khả năng phân hủy diazinon từ các mẫu đất bằng phương pháp làm giàu mật số vi khuẩn, trong môi trường khoáng tối thiểu có bổ sung diazinon đạt nồng độ 20 ppm và tách ròng trên môi trường TSA. Xác định khả năng phân hủy diazinon của các dòng vi khuẩn bằng phương pháp theo dõi hàm lượng diazinon còn lại sau 30 ngày nuôi ủ. Các dòng vi khuẩn phân hủy diazion, được định danh dựa vào trình tự gen 16Sr RNA so sánh với các trình tự nucleotide của các chủng vi khuẩn trên NCBI, xây dựng cây phả hệ mối quan hệ di truyền của các dòng vi khuẩn bằng phương pháp Maximun-Likelihood. Sự thay đổi của cấu trúc cộng đồng vi khuẩn phân hủy diazinon, được xác định bằng kỹ thuật điện di biến tính tăng cấp (DGGE).

Trên mô hình chuyên màu, tổng số có 21 mẫu đất được thu tại một số địa điểm ở Đồng bằng sông Cửu Long. Kết quả bố trí thí nghiệm cho thấy có 10 cộng đồng vi khuẩn có khả năng phân hủy diazinon hiệu quả, giảm từ 14,3% đến 37,9% sau 14 nuôi ủ trong môi trường khoáng tối thiểu. Kết quả phân lập được 87 dòng vi khuẩn, trong đó có 15 dòng vi khuẩn có khả năng phát triển trong môi trường khoáng tối thiểu có bổ sung diazinon có nồng độ 20 ppm. Tuy nhiên, chỉ có 4 dòng vi khuẩn ký hiệu HA7.4, TA3.2, TA4.17 và HA7.1 có khả năng phân hủy thuốc trừ sâu diazinon hiệu quả, giảm từ 15,4% đến 27,9% sau 30 ngày nuôi cấy. Mật số của bốn dòng vi khuẩn này đạt cao nhất trong điều kiện môi trường nuôi cấy như sau: ở 30oC, pH 7, có bổ sung thêm nguồn TSB và ở nồng độ diazinon dao động từ 20 ppm–50 ppm. Các dòng vi khuẩn thể hiện khả năng phân hủy diazinon hiệu quả nhất ở nhiệt độ 30oC với tốc độ phân hủy dao động từ 0,55-0,94%/ngày, pH 6-7 với tốc độ phân hủy từ 0,50-0,94%/ngày, ở nồng độ Diazinon 20 ppm với tốc độ phân hủy từ 0,56-0,93%/ngày và với mật số vi khuẩn ban đầu đạt 106 CFU/mL có tốc độ phân hủy 0,60-0,98%/ngày.

Trên mô hình canh tác luân canh lúa–màu, tổng số có 20 mẫu đất được thu tại một số địa điểm ở Đồng bằng sông Cửu Long. Kết quả nghiên cứu cho thấy có 13 cộng đồng vi khuẩn thể hiện khả năng phân hủy diazinon, giảm từ 17,6% đến 97,8% sau 14 ngày nuôi cấy. Bên cạnh đó, 27 trong tổng số 109 chủng vi khuẩn phân lập từ 13 cộng đồng vi khuẩn phát triển tốt trong môi trường khoáng tối thiểu có bổ sung diazinon đạt nồng độ 20 ppm. Tuy nhiên, chỉ có 2 dòng vi khuẩn ký hiệu CL36_M4 và BT4_L1 phân hủy hiệu quả diazinon lần lượt là 20,9% và 29,3% sau 30 ngày nuôi cấy. Mật số của hai dòng vi khuẩn CL36_M4 và BT4_L1 đạt cao nhất ở điều kiện môi trường như: 30oC, pH 7, môi trường nuôi cấy bổ sung TSB và nồng độ diazinon từ 20–50 ppm. Hai dòng vi khuẩn CL36_M4 và BT4_L1, phân hủy diazinon hiệu quả nhất ở điều kiện môi trường như 30oC với tốc độ phân hủy từ 0,70-1,03 %/ngày, pH 7 với tốc độ phân hủy dao động từ 0,72-1,02 %/ngày, ở nồng độ diazinon 20 ppm với tốc độ phân hủy từ 0,67-0,97 %/ngày và với mật số vi khuẩn chủng vào ban đầu ở 106 CFU/mL cho tốc độ phân hủy từ 0,74-1,18 %/ngày.

Dòng vi khuẩn HA7.1 thể hiện khả năng phân hủy diazinon hiệu quả trong môi trường đất dưới điều kiện phòng thí nghiệm sau 30 ngày nuôi ủ. Các nghiệm thức có bổ sung vi khuẩn HA7.1 làm giảm hàm lượng diazinon và khác biệt có ý nghĩa thống kê so với nghiệm thức đối chứng. Trong đó, nghiệm thức 4: Đất không tiệt trùng và bổ sung vi khuẩn HA7.1 có hàm lượng diazinon giảm mạnh hơn so với nghiệm thức 2: Đất tiệt trùng và bổ sung vi khuẩn HA7.1, giảm tương ứng là 63,7% với tốc độ phân hủy diazinon 2,11%/ngày và 48,6% với tốc độ 1,62%/ngày.

Trên mô hình luân canh lúa màu ngoài đồng ruộng, cấu trúc của cộng đồng vi khuẩn hiếu khí phân hủy diazinon trong đất vào vụ màu chứa 3 nhóm vi khuẩn khác nhau, trong khi vụ lúa, chỉ chứa 2 nhóm vi khuẩn. Trong điều kiện nhà lưới, trên mô hình chuyên màu cấu trúc của cộng đồng vi khuẩn phân hủy diazinon trong đất được phân tách thành 6 nhóm vi khuẩn khác nhau. Trong trong mô hình luân canh lúa-màu cho thấy đất vụ màu có chứa 4 nhóm vi khuẩn khác nhau, trong khi đất trong vụ lúa chứa 3 nhóm vi khuẩn khác nhau.

Kết quả giải mã trình tự gen 16S rRNA của sáu dòng vi khuẩn HA7.1, HA7.4, TA3.2, TA4.17, CL36_M4 và BT4_L1 cho thấy chúng lần lượt được định danh như là Lactobacillus plantarum HA7.1, Pandoraea sp. HA7.4, Sinomonas atrocyanea TA3.2, Achromobacter xylosoxidans TA4.17, Cupriavidus sp.CL36_M4 và Achromobacter xylosoxidans BT4_L1.

  1. Những kết quả mới của luận án

- Kết quả phân lập được 196 dòng vi khuẩn có khả năng phân hủy thuốc trừ sâu Diazinon có nguồn gốc từ đất canh tác luân canh lúa-màu và chuyên màu ở 4 tỉnh ĐBSCL.

- Ứng dụng phương pháp theo dõi hàm lượng thuốc trừ sâu Diazinon còn lại trong môi trường khoáng tối thiểu sau khi chủng vi khuẩn 30 ngày, kết quả đã xác định được 6 dòng vi khuẩn phân hủy Diazinon hiệu quả cao được phân lập từ mô hình canh tác luân canh lúa-màu và chuyên màu.

- Xác định được một số nhân tố môi trường (nhiệt độ, pH, nguồn cacbon, nồng độ Diazinon) thúc đẩy cho sự gia tăng mật số và phân hủy Diazinon của 6 chủng vi khuẩn (Lactobacillus plantarum HA7.1, Pandoraea sp. HA7.4, Sinomonas atrocyanea TA3.2,  Achromobacter xylosoxidans TA4.17, Cupriavidus sp.CL36_M4 và Achromobacter xylosoxidans BT4_L1) trong môi trường khoáng tối thiểu.

  1. Các ứng dụng/ khả năng ứng dụng trong thực tiễn, các vấn đề cần tiếp tục nghiên cứu:

- Có thể ứng dụng các chủng vi khuẩn Lactobacillus plantarum HA7.1, Pandoraea sp. HA7.4, Sinomonas atrocyanea TA3.2,  Achromobacter xylosoxidans TA4.17, Cupriavidus sp.CL36_M4 và Achromobacter xylosoxidans BT4_L1 trong xử lý ô nhiễm hoạt chất thuốc trừ sâu Diazinon.

- Kết quả nghiên cứu của đề tài làm nguồn tư liệu quí báu cho các nghiên cứu chuyên sâu trong tương lai. Các dòng vi khuẩn có khả năng phân hủy hoạt chất thuốc trừ sâu Diazinon hiệu quả có thể ứng dụng giảng dạy trong lĩnh vực xử lý ô nhiễm môi trường.

  1. Research Abstract

Organic agricultural pesticides are so stable in ecosystems and biosphere. These compounds affect human health and envonrinment. Diazinon, a toxic  substance, has  been banned or restricted in most countries. However, some places farmers still continue using this substance in their farm as a pesticide to control the pest. Diazinon degrading bacteria have been extensively studied in temperate soils, but information on behaviour in tropical soils is limited. The aim of this study was(1) to isolate and characterize  diazinon degrading bacteria from upland crop-rice and continuous upland cropping system soils in the Mekong Delta of Vietnam. (2) to specify the effects of temperature, pH, carbon sources and diazinon concentration on the growth of some selected bacterial isolates, and to find out the optimal conditions of innoculation environment uch as temperature, pH, innoculation density and diazinon concentration to enhance the biodegradation rate of diazinon. (3) to evaluate the effects of two different cropping systems (continuous upland crop and  upland crop-rice) on diazinon degradation under nethouse condition and in the actual fields.(4) to test the effects of different cropping systems on the diazinon degrading bacterial community structure under nethouse condition.

The isolation of diazinon degrading bacteria was performed with soil collected from continuous upland crop and  upland crop-rice cropping systems. Soil bacteria were enriched in mineral salt medium solution containing 20 ppm diazinon as the only carbon source for bacterial growth. Tryptic Soya Agar (TSA) was used for the step of purification of bacterial strains. Diazinon degrading bacteria were identified based on the sequencing of 16S rRNA gene, and these sequences were aligned with published sequences in NCBI BLAST nucleotide database. Then phylogenetic trees were established based on the 16S rRNA sequences by the Maximum-Likelihood method. The diazinon degrading bacterial community structure analysis was examined by PCR-denaturing gradient gel electrophoresis (DGGE). 

Twenty one soil samples were collected from continuous upland crop cropping system in the Mekong Delta of Vietnam. Results showed that 10 out of 21 bacterial communities showed their degradation capacity toward diazinon from 14.3% to 37.9% of the initially applied diazinon concentration within 14 incubation days and percentage of the remained diazinon in the liquid culture of these communities was significantly lower than that of the control treatment. Fifteen out of 87 bacterial strains which were isolated from these 6 bacterial communities showed a well growing capacity through the turbidity index in minimal salt medium containing 20 ppm of diazinon. Four bacterial strains which were coded namely as HA7.4, TA3.2, TA4.17 and HA7.1 showed efficiently in degradation of diazinon. The degradation capacity of these single strains varied from 15.4% to 27.9% of the initially applied diazinon concentration in 30 incubation days and was significantly different from the control treatment.

The experiments were established under the laboratory conditions to test the effects of environmental conditions such as temperature, pH and carbon source on the growth of some selected strains in minimal salt medium in 5 days. Results showed that bacterial cell numbers were obtained at the highest numbers when grown under the conditions of 30oC, pH 7 of the medium (except the strain HA7.1 at pH 6), tryptic soya broth (TSB) as a carbon source and diazinon concentration of 20 ppm-50 ppm. The tested bacterial strains showed their higher degradation of diazinon when environmental conditions was at 30oC (the degradation rate of 0,55-0,94% day-1), pH 6-7 (the degradation rate of 0,50-0,94% day-1), the diazinon concentration of 20 ppm (the degradation rate of 0,56-0,93% day-1) and the inintial innoculation density of 106 CFU/mL (the degradation rate of 0,60-0,98% day-1).

Twenty soil sample were collected  from upland crop-rice cropping systems in the Mekong Delta of Vietnam. The results showed that 13 out of 20 bacterial communities showed their degrading capacity toward diazinon. Degradation of diazinon by these bacterial communities varied between 17,6% and 97,8% after 14 day of incubation. However, only 2 bacterial strains revealed good degradation for diazinon, namely CL36_M4 and BT4_L1 and their degradation capacity was 20,9% and 29,3% of the initial applied concentration after 30 days of incubation, respectively. Two strains CL36_M4 and BT4_L1 achieved the highest cell numbers when grown under conditions of 30oC, pH 7 of the medium, TSB as a carbon source and diazinon concentration of 20-50 ppm. These two bacterial strains showed their most effectiveness in degradation of diazinon when grown under the environmental conditions of 30oC (the degradation rate of 0,70-1,03% day-1), pH 7 (the degradation rate 0,72-1,02% day-1), the diazinon concentrations of 20 ppm (the degradation rate of 0,67-0,97% day-1) and the inintial innocuation density of 106 CFU/mL (the degradation rate of 0,74-1,18% day-1).

The bacterial strains HA7.1 showed its better degradation of diazinon in soil, under the laboratory conditions within 30 incubation days. The concentration of diazinon was reduced in treatments inoculated this strain and significantly higher than the control treatment. Treatments of non-sterilized soil in combination with HA7.1 innoculation degraded more diazinon than that of sterilized soil, about 63,7% of the initially applied concentration (the degradation rate of 2,11% day-1) and 48,6% (the degradation rate of 1,62% day-1), respectively.

According to the numbers of bands showed on the profice of DGGE gels the diazinon degrading bacterial community structure in upland crop cropping system had 3 bands whereas the continueous rice cropping system had only two bands. It means that the bacterial communities in the upland crop cropping system was more diversified than that of the continueous rice cropping system and similarly under the nethouse condition, the continuous upland crop cropping systems had six bands on the DGGE gel while the DGGE gel profiles of soil bacterial communities in the upland-rice crop rotation cropping system showed four bands and three bands for upland crop soil samples and rice crop soil samples, respectively. It means also that the bacterial communities in the upland crop cropping system was more diversified than that of the upland crop-rice rotation cropping systems.

According to 16s RNA gene sequencing of six diazinon degrading bacterial strains, coded namely as HA7.1, HA7.4, TA3.2, TA4.17, CL36_M4 and BT4_L1, were identified as species of Lactobacillus plantarum HA7.1, Pandoraea sp. HA7.4, Sinomonas atrocyanea TA3.2,  Achromobacter xylosoxidans TA4.17, Cupriavidus sp.CL36_M4 and Achromobacter xylosoxidans BT4_L1, respectively.

  1. Research Creativeness

One hundred ninety six bacterial strains can degrade Diazinon which were isolated from upland crop-rice and continuous upland crop farming system soils in four provinces in the Mekong Delta region of Vietnam.

The results is determined that have six bacterial strains revealed good degradation for Diazinon by diazinon concentrate tracking mothod in mineral salt medium solution after 30 incubation  days.

Determine the effects of environmental conditions on the bacterial growth  and Diazinon biodegradation rate of six bacterial strains (Lactobacillus plantarum HA7.1, Pandoraea sp. HA7.4, Sinomonas atrocyanea TA3.2,  Achromobacter xylosoxidans TA4.17, Cupriavidus sp.CL36_M4 and Achromobacter xylosoxidans BT4_L1)  revealed good degradation in mineral salt medium solution.

  1. Practical implications from study

Six bacterial strains such as Lactobacillus plantarum HA7.1, Pandoraea sp. HA7.4, Sinomonas atrocyanea TA3.2,  Achromobacter xylosoxidans TA4.17, Cupriavidus sp.CL36_M4 and Achromobacter xylosoxidans BT4_L1 can use to treat Diazinon pesticide pollution. The results of this thesis were used as resources for scientific researching in the future. The bacterial strains strains showed the most effective in degradation of Diazinon can use as resources for teaching about environmental pollution treatment.
