This study was conducted at Can Tho University from 2017 to 2025. The main objective of the research is to assess the effects of salinity on the structure of the plankton community in the lower Hau River region, thereby providing essential data for forecasting and proposing solutions to mitigate the adverse impacts of saline intrusion on aquaculture. The research was implemented through the collection and analysis of plankton samples from the Hau River (including Cai Con, Dai Ngai, and Tran De sites), combined with two experimental systems designed to simulate saltwater intrusion conditions. The main findings of the research are summarized as follows:
Regarding the water quality of the Hau River, ten key water quality indicators were measured at the river estuary, including temperature (29.2±1.07°C), pH (7.4±0.49), dissolved oxygen (DO, 5±1.77 mg/L), chemical oxygen demand (COD, 13.5±10.58 mg/L), biochemical oxygen demand (BOD₅, 3.5±1.24 mg/L), nitrate (NO₃-, 0.4±0.32 mg/L), total ammonia nitrogen (TAN, 0.1±0.06 mg/L), total nitrogen (TN, 1.8±1.06 mg/L), phosphate (PO₄³-, 0.3±0.19 mg/L), and total phosphorus (TP, 0.9±0.44 mg/L). Overall, these parameters were within acceptable ranges for aquaculture activities during the sampling period. Salinity levels in the river fluctuated between 0 and 20‰, with average values of 0.13±0.448‰ at Cai Con, 1.04±1.829‰ at Dai Ngai, and 5.71±6.610‰ at Tran De. Strong saline intrusion was observed in the downstream area of the Hau River from December 2017 to April 2018.
The phytoplankton community in the downstream area of the Hau River comprised 221 species belonging to five phyla. Among these, Bacillariophyta was the dominant group with 97 species (43.9%) and an average density of 45,063±31,263 cells/L, followed by Chlorophyta with 54 species (24.4%, 12,489±8,291 cells/L), Euglenophyta with 35 species (15.8%, 6,653±3,958 cells/L), Cyanobacteria with 19 species (8.6%, 5,393±3,330 cells/L), and Dinophyta with 16 species (7.2%, 4,154±4,113 cells/L). Correlation analysis revealed that the total number of phytoplankton species was negatively correlated with salinity (r=-0.631, p=0.016), whereas total phytoplankton density showed a positive correlation with salinity (r=0.247, p=0.000). Furthermore, the number of species within the phyla Cyanobacteria (r=-0.378, p=0.001), Chlorophyta (r=-0.378, p=0.001), Euglenophyta (r=-0.359, p=0.002), and Bacillariophyta (r=-0.608, p=0.021) were all negatively correlated with salinity. In contrast, the density of Bacillariophyta was positively correlated with salinity (r=0.421, p=0.000). Notably, the phylum Dinophyta exhibited a positive correlation with salinity in both species richness and density (r=0.644, p=0.000; r=0.718, p=0.000, respectively). The occurrence of saltwater intrusion in the lower Hau River thus resulted in an increase in the density of Bacillariophyta, which is considered beneficial for aquaculture; however, the concurrent rise in Dinophyta abundance represents a potential negative impact.
The zooplankton community in the downstream area of the Hau River consisted of 137 species classified into five major groups. Rotifera was the most dominant group with 47 species (34%) and an average density of 19,741±17,485 individuals/m³, followed by Copepoda with 44 species (32%, 19,920±13,674 individuals/m³), Protozoa with 26 species (19%, 6,811±5,040 individuals/m³), Cladocera with 12 species (9%, 4,876±4,002 individuals/m³), and eight other taxa representing 6% with an average density of 10,871±8,926 individuals/m³. Correlation analysis indicated that the total number of zooplankton species in the Hau River was negatively correlated with salinity (r=-0.300, p=0.010). In contrast, both the number of species and the density of Protozoa (r=0.439, p=0.000; r=0.497, p=0.000, respectively) and Copepoda (r=0.457, p=0.000; r=0.458, p=0.000, respectively) showed positive correlations with salinity. Conversely, the number of Rotifera (r=-0.690, p=0.003) and Cladocera (r=-0.459, p=0.000) species, as well as their densities (Rotifera: r=-0.282, p=0.025; Cladocera: r=-0.294, p=0.012), exhibited negative correlations with salinity. These findings suggest that even mild saline intrusion can adversely affect the natural food sources of zooplankton and, consequently, freshwater aquaculture activities along the Hau River.
In the two experimental systems simulating salinity intrusion-one driven by tidal influence (Experiment 1) and the other by duration (Experiment 2)-a total of 109 and 186 phytoplankton species were recorded, respectively. In Experiment 1, Chlorophyta was the dominant group with 45 species (41%) and an average density of 45,586±7,015 cells/L, followed by Euglenophyta with 39 species (36%, 20,762±2,843 cells/L), Bacillariophyta with 17 species (16%, 84,062±11,545 cells/L), and Cyanobacteria with 8 species (7%, 25,120±1,662 cells/L). In Experiment 2, Chlorophyta again dominated with 80 species (43%, 87,063±11,843 cells/L), followed by Euglenophyta with 50 species (27%, 15,517±2,313 cells/L), Bacillariophyta with 44 species (24%, 99,388±16,463 cells/L), and Cyanobacteria with 12 species (6%, 14,959±2,184 cells/L). Correlation analysis revealed that salinity had a significant effect (p<0.05) on phytoplankton community structure. In both experiments, the total number of phytoplankton species (r₁=-0.549, p₁=0.000; r₂=-0.405, p₂=0.001) and total phytoplankton density (r₁=-0.307, p₁=0.000; r₂=-0.163, p₂=0.024) were negatively correlated with salinity. The high abundance of Bacillariophyta and the broad distribution of Cyanobacteria observed in the salinity intrusion simulations may serve as useful biological indicators for predicting both the potential benefits and drawbacks to aquaculture activities in the downstream areas of the Hau River during saltwater intrusion events.
For zooplankton, the study identified 52 species in Experiment 1 (simulating salinity intrusion by tidal influence) and 92 species in Experiment 2 (simulating salinity intrusion over time). In Experiment 1, Rotifera was the dominant group with 22 species (42.3%) and an average density of 2,275,758±248,768 individuals/m³, followed by Copepoda with 12 species (23.1%, 87,792±5,984 individuals/m³), Protozoa with 11 species (21.2%, 378,089±60,024 individuals/m³), Cladocera with 4 species (7.7%, 66,792±13,502 individuals/m³), and three other taxa accounting for 8% with an average density of 12,263±1,365 individuals/m³. In Experiment 2, Rotifera remained dominant with 37 species (40.2%) and an average density of 2,866,514±241,077 individuals/m³, followed by Copepoda with 16 species (17.4%, 134,802±9,096 individuals/m³), Protozoa with 30 species (32.6%, 313,761±17,582 individuals/m³), Cladocera with 6 species (6.5%, 36,476±7,165 individuals/m³), and three other taxa representing 3.3% with an average density of 4,476±785 individuals/m³. Correlation analysis indicated that salinity had a significant effect (p<0.05) on zooplankton community composition. The total number of zooplankton species showed a negative correlation with salinity in both experiments (r₁=-0.331, p₁=0.007; r₂=-0.563, p₂=0.000). The observed reduction in zooplankton species richness under simulated salinity intrusion conditions suggests that salinity fluctuations could negatively affect natural food resources and the biological productivity of aquatic ecosystems in the downstream areas of the Hau River.
The dissertation established a dataset on the community structure of plankton along a salinity gradient in the downstream area of the Hau River in the Mekong Delta, where no previous studies had been conducted. A total of 221 phytoplankton species and 137 zooplankton species were recorded. The total numbers of both phytoplankton and zooplankton species were negatively correlated with salinity, while the results also demonstrated differential responses among plankton groups to salinity fluctuations in the Hau River.
The dissertation further elucidated the differential responses of plankton groups of significance to aquaculture under the influence of salinity. The species richness and abundance of Rotifera and Cladocera decreased, whereas Dinophyta exhibited a marked increase in both species richness and abundance. These findings enable the identification of changes in natural food resources and the potential proliferation of harmful algal groups under saline intrusion in the downstream Hau River. Accordingly, the study contributes additional baseline data for assessing the impacts of saline intrusion on aquaculture in the Mekong Delta.
The dissertation also provided experimental evidence from two experiments simulating saline intrusion under tidal cycles characteristic of the Mekong Delta, thereby contributing to the validation of the effects of salinity on plankton community structure. The results demonstrated that salinity significantly affected the community structures of both phytoplankton and zooplankton (p < 0.05), with the total number of species in both groups showing a decreasing trend as salinity increased. These findings provide further evidence of changes in plankton communities in response to saline intrusion and establish an experimental basis for forecasting and developing measures to enhance the adaptive capacity of aquaculture to saline intrusion under the increasingly pronounced impacts of climate change in the Mekong Delta, both in recent years and in the future.
Practical applications
The findings of this dissertation provide an empirical database on changes in plankton communities under the influence of saline intrusion and establish regression models with practical value for identifying and predicting changes in natural food resources, particularly Rotifera and Cladocera. The models also provide an early warning of the potential proliferation of potentially harmful dinoflagellate groups (Dinophyta) in aquaculture areas affected by saline intrusion. These findings have important practical implications by translating ecological changes into indicators that can support management and decision-making in aquaculture. They provide a valuable scientific basis for management authorities to proactively plan and zone aquaculture areas according to salinity conditions, adjust farming calendars, select appropriate cultured species, and develop water quality management strategies, particularly for extensive aquaculture systems.
Furthermore, the findings of this dissertation provide data on the responses of plankton groups to saline intrusion, thereby supporting managers and aquaculture farmers in restructuring cultured species according to salinity gradients. This may involve prioritizing the cultivation of marine species or species with high salinity tolerance in areas experiencing increased salinity during the dry season. Such an approach can contribute to enhancing the preparedness, efficiency, and adaptive capacity of aquaculture in the downstream area of Hau River in response to the increasingly complex impacts of saline intrusion and climate change in the Mekong Delta.
Recommendations for future research
Future research should further expand the scope and enhance the ecological relevance of saline intrusion simulation experiments by designing studies based on characteristic tidal cycles, with monitoring periods extending from three to six months. These experiments should use water and sediment samples collected directly from the Hau River and investigate different levels and rates of salinity increase in order to more accurately reflect the dynamics of saline intrusion under natural conditions.
In addition, future studies should focus on quantifying the effects of salinity fluctuations on the entire natural food web and clarifying the mechanisms and extent to which declines in Rotifera and Cladocera affect the overall availability and quality of natural food resources for cultured species. Further research is also needed to comprehensively assess the risks associated with the occurrence and proliferation of harmful algal bloom (HAB) groups and their potential impacts on aquaculture activities.
In particular, integrated studies should be conducted to examine the relationships among changes in plankton community structure, the quality of natural food resources, growth performance, survival, and productivity across specific cultured species and aquaculture systems in the downstream area of Hau River. Such research would provide a stronger scientific basis for developing predictive tools and adaptive management strategies to support sustainable aquaculture under increasingly severe saline intrusion.
