  1. Content of thesis summary

- Regarding the morphological species composition:

Based on histological analyses of the male pores and copulatory pouches, this dissertation contributes to clarifying the structural characteristics and taxonomic significance of the copulatory pouch, a structure that has been the subject of considerable debate and differing interpretations in earthworm taxonomy: The copulatory pouch can be defined as a pouch-like structure with a distinct muscular layer, formed by the inward invagination of the body wall surrounding the male pore, either with or without folds, thereby creating a relatively complex internal cavity. The copulatory pouch is located at the distal part of the male reproductive system and constitutes an external chamber associated with the male pore. It may be eversible and may contain accessory papillae, accessory glands, a penis, copulatory setae, or micronephridia. And elucidate the relationship between the male pore and the copulatory pouch: it can be seen from the analysis that the superficial male pore has the primary male pore directly open onto the body surface; the vertical slit-like male opening has the primary male pore roofed by the overgrowth of the body wall’s external layers; while the transverse slit-like or round male openings can have the primary male pore open either into an inner-smooth surface or an inner-folds surface copulatory pouches.

Based on the histological structure of the copulatory pouch and the original species descriptions, the thesis proposed the taxonomic reassignment of nine species from Metaphire to Amynthas: Amynthas fluvialoides, Amynthas munglongmontis, Amynthas stephensoni, Amynthas thaibinhensis, Amynthas decemtheca, Amynthas giengensis, Amynthas mochauana, Amynthas posthuma, and Amynthas varians songbana. In addition, with the description of two new species, Metaphire mangcanhensis and Metaphire dalatensis, the thesis determined that 64 species and subspecies of Metaphire are currently recorded in Vietnam.

The thesis identified: Metaphire bahli has two morphological forms based on the shape of the male region and state of genital markings while Metaphire peguana has two subspecies, Metaphire peguana peguana, and Metaphire peguana laisonensis. Metaphire peguana peguana had genital markings disc-shaped in 17/18/19 and a diverticulum attached to the base of the ampulla duct while Metaphire peguana laisonensis had genital papillae slit-like shaped in xvii and xix, and diverticulum end at the middle of ampulla duct.

- Regarding the molecular species composition:

Regarding K2P genetic distances, for the COI gene, intraspecific genetic distances ranged from 0–11.3%, lower than interspecific genetic distances (14.3–24.5%). For the 16S-rRNA gene, intraspecific genetic distances ranged from 0–4.4%, whereas interspecific genetic distances ranged from 4.4–14.3%.

During the molecular analyses, ABGD analyses of the COI, 16S-rRNA, and COI-16S datasets separated samples of Metaphire easupana, Metaphire pacseana, and Metaphire neoexilis into different MOTUs. However, these delimitations were not supported by the phylogenetic tree analyses. Based on phylogenetic evidence, there was insufficient support for recognizing these MOTUs as independent putative species; therefore, the samples were retained within the same MOTU as their corresponding morphospecies. Based on the combined results from the three datasets (COI, 16S-rRNA, and COI-16S), the study identified 27 molecular species in the sampled material, corresponding exactly to the 27 morphospecies recognized, some of which exhibited high levels of genetic divergence.

- Regarding the taxonomic position of the genus Metaphire within the Pheretima group in Vietnam: The thesis continues to recognize Metaphire as an independent genus within the caecate group of Pheretima sensu lato Kinberg, 1867. The copulatory pouch remains an important morphological character for distinguishing Metaphire from Amynthas.

  1. The novel contributions of the thesis

- Regarding the specimen collection: This dissertation added 669 ethanol-preserved specimens representing 21 species, including six species that were previously unrepresented in the collection. In addition, 35 formalin-preserved specimens representing six species were added, including one species that had not previously been represented. The taxonomic status of nine species was revised by transferring them from the genus Metaphire to the genus Amynthas. Furthermore, two species new to science, Metaphire mangcanhensis Phan, Lam & Nguyen, 2025 và Metaphire dalatensis Phan, Lam & Nguyen, 2025, were described.

- Regarding histological slides: This dissertation added 98 transverse histological sections of the male pore region representing 97 species within the Pheretima complex, including 55 slides representing 55 species of Amynthas, one slide representing one species of Metapheretima, 35 slides representing 34 species of Metaphire (M. bahli was represented by two slides), one slide representing one species of Pheretima, four slides representing four species of Planapheretima, and two slides representing two species of Polypheretima.

- Regarding molecular data: This dissertation deposited 58 COI sequences and 80 mitochondrial 16S-rRNA sequences of Metaphire species in GenBank.

- Regarding the taxonomic position of the genus Metaphire: This is the first study in Vietnam to integrate morphological and molecular data to investigate the taxonomic position of the genus Metaphire within the Pheretima group and to compile and systematize data on the species diversity of this genus in Vietnam.

  1. Applications and Directions for future research

- The findings on the species composition of the genus Metaphire will contribute to a more comprehensive understanding of biodiversity in Vietnam. The thesis also provides additional data for analyzing phylogenetic relationships among genera within the Pheretima group, thereby contributing to the taxonomic study of earthworms in Vietnam.

- Further specimen collection and expanded analyses of species belonging to the genus Metaphire and other genera within the Pheretima group are needed to provide further insights into the diversity and taxonomic positions of this group.

- Expand the integrative approach by combining morphological data with molecular data from multiple markers to further clarify phylogenetic relationships, taxonomic boundaries, and the taxonomic significance of morphological characters within the Pheretima group.
