The objectives of this study were to establish a scientific basis and develop a technological process for producing a soluble dietary fiber-rich preparation from citrus fruit by-products, as well as to evaluate its structural characteristics, functional properties, and potential applications in food products.
Statistical data indicate that the Mekong Delta has the largest citrus-growing area and production volume in the country. Five commonly cultivated citrus fruits were investigated: Da Xanh pomelo, Nam Roi pomelo, Sanh orange, sweet orange, and seedless limes. The results showed that the albedo and pulp residues, including the core and segment membranes, of Sanh orange contained a high level of total dietary fiber (TDF) and a relatively high proportion of soluble dietary fiber (SDF). These by-products are therefore considered suitable for the technological objective of producing an SDF-rich dietary fiber preparation for food applications. Sanh orange by-products were pretreated with 96% ethanol at a solid-to-solvent ratio of 1:3 g/mL at 60°C for 30 min. This treatment improved the soluble fiber content and removed soluble components that could adversely affect the sensory quality of the resulting product.
However, the major challenge of this study was maximizing the release of soluble fiber from the raw material. Therefore, individual extraction methods using citric acid, alkali, and enzymes, as well as combined methods assisted by microwave and ultrasound treatments, were investigated in this study. Following the screening process, cellulase treatment combined with microwave-assisted citric acid extraction demonstrated superior effectiveness. This innovative combination partially disrupted and modified the insoluble fiber structure, resulting in an extraction yield of 48.53%, with soluble fiber accounting for 63.29% of the preparation.
Following the establishment of the technological process, the structural characteristics of the SDF-rich dietary fiber preparation obtained from Sanh orange by-products were elucidated using modern analytical techniques, including Fourier-transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), and scanning electron microscopy (SEM). Microstructural observations revealed that the fiber preparation exhibited a porous structure, high hydrophilicity, and an improved hydration capacity. As a result of these favorable microstructural modifications, the physiological functional properties of the material were also substantially enhanced. Specifically, the water-holding capacity, oil-holding capacity, and swelling capacity were 28.94 g/g, 5.82 g/g, and 17.26 mL/g, respectively. Notably, following purification by dialysis, the preparation contained high levels of soluble fiber and pectin and exhibited properties comparable to those of commercial dietary fibers.
In parallel with the evaluation of physicochemical properties, the biological activities of the dietary fiber preparations were also examined. The prepared material exhibited a good glucose adsorption capacity, ranging from 0.88 to 8.58 mmol/g, which was comparable to that of the commercial sample. However, its antioxidant activity was relatively low, with IC₅₀ values determined using the DPPH and phosphomolybdenum assays of 1,252.45 and 8,551.67 µg/mL, respectively. The corresponding values were 686.12 and 6,030 µg/mL for the commercial sample and 71.69 and 143.15 µg/mL for Trolox control. Additionally, the preparation exhibited prebiotic activity by promoting the growth of the probiotic strains Lactiplantibacillus plantarum and Bifidobacterium animalis subsp. lactis, with prebiotic indices of 2.16 and 2.38, respectively. Finally, acute toxicity testing demonstrated that the obtained dietary fiber preparation was safe under the conditions investigated.
To demonstrate its practical value, dietary fiber preparation derived from Sanh orange by-products was incorporated into red tilapia fish cake. Its performance was compared with that of several commonly used commercial dietary fibers, including soybean, wheat, citrus, and commercial orange fibers. The results indicated that dietary fiber supplementation did not significantly affect the moisture content, water-holding capacity, lipid content, or product whiteness. At a supplementation level of 0.75%, the dietary fiber powder derived from Sanh orange by-products maintained the gel structure of red tilapia fish cake, showing an effect comparable to that of soybean fiber. At this ratio, the product achieved a high gel strength of 986.72 ± 28.03 g·cm and a water-holding capacity of 98.77 ± 0.38%, while maintaining desirable sensory quality and enhancing its nutritional value.
The study contributes to the value-added utilization of citrus fruit by-products in the Mekong Delta through the development of functional food ingredients, in line with circular economy principles and sustainable development. Dietary fiber recovered from Sanh orange by-products has potential as a fiber-enriching ingredient, enhancing the utilization of these by-products and supporting the development of value-added food products.
Further research directions: Investigate the stability of the recovered dietary fiber and appropriate preservation and storage methods; extend its applications to other food products; and examine ultrafiltration combined with tangential-flow filtration or multistage counter-current washing to facilitate the scale-up of the purification process.
