  1. Tóm tắt nội dung luận án

Luận án bao gồm 5 nội dung nhằm xác định tình hình nhiễm sán dây trên chó tại một số tỉnh ở Đồng bằng sông Cửu Long (ĐBSCL), xác định thành phần loài sán dây dang lưu hành trên chó tại các tỉnh ĐBSCL, nghiên cứu xác định vòng đời phát triển của loài sán dây Dipylidium caninum. Ngoài ra đề tài sẽ góp phần hoàn thiện mô tả triệu chứng lâm sàng và khảo sát bệnh tích đại thể, vi thể và thử nghiệm thuốc tẩy trừ sán dây trên chó.

Kết quả nghiên cứu cho thấy:

Qua phương pháp kiểm tra phân và mổ khám tìm sự hiện diện sán dây trên chó tại 6 tỉnh ĐBSCL (Cần Thơ, An Giang, Đồng Tháp, Kiên Giang, Sóc Trăng, Bến Tre) từ 2014 đến 2018 cho thấy, chó nhiễm sán dây với tỷ lệ nhiễm chung là 22,51% (đối với phương pháp kiểm tra phân) và 25,86% (đối với phương pháp mổ khám), với 5 loài sán dây được tìm thấy thuộc 2 bộ Cyclophyllidea Pseudophyllidea Dipylidium caninum, Taenia pisiformis, Taenia hydatigena, Spirometra mansoni, Diphyllobothrium latum.

Phân tích loài Dipylidium caninum qua đặc điểm hình thái và kỹ thuật sinh học phân tử qua khuếch đại vùng gene ITS và gene 28S. Nghiên cứu đối chiếu trình tự của nucleotide của đoạn gene ITS thu được từ mẫu cùng đoạn gene này trong GenBank với 99% tương đồng về kiểu gene sán dây trong nghiên cứu so với mẫu sán đã được đăng ký trong GenBank.

Kết quả nghiên cứu xác định được thời gian hoàn thành vòng đời sán dây Dipylidium caninum trải qua 2 giai đoạn: Bên trong ký chủ trung gian Ctenocephalides và bên trong ký chủ chính (chó).

Nghiên cứu bệnh lý ở chó nhiễm sán dây thể hiện triệu chứng lâm sàng phổ biến là ngứa cắn hậu môn, tiêu chảy đồng thời chó bệnh sán dây thường xuất hiện bệnh tích phổ biến nhất là xuất huyết cục bộ tại vị trí bám của đầu sán. Bệnh tích vi thể trên hệ tiêu hóa chó nhiễm sán dây Dipylidium caninum thể hiện tổn thương trên niêm mạc ruột, cơ ruột và hạch bạch huyết ruột với dấu hiệu điển hình như đầu cầu lông nhung bong tróc, nhiều bạch cầu hiện diện trên niêm mạc ruột và áo cơ, viêm cơ và áo cơ ruột, viêm sợi huyết trên bề mặt niêm mạc, tăng sinh hạch bạch huyết.

Thuốc praziquantel và niclosamide có hiệu quả điều trị rất cao trong điều trị bệnh sán dây trên chó với tỷ lệ sạch sán 100% ở liều 10 mg/kg thể trọng đối với praziquantel và 150 mg/kg thể trọng đối với niclosamide.

  1. Những đóng góp mới của luận án

Đã xác định các đặc điểm dịch tễ học bệnh sán dây trên chó và xác định một số yếu tố nguy cơ liên quan đến tỷ lệ nhiễm sán dây ký sinh, lượng hóa các yếu tố nguy cơ liên quan đến tỷ lệ nhiễm sán dây trên chó bằng tỷ suất chênh (Odd ratio).

Đã xác định được đoạn mồi nhận diện loài sán dây Dipylidium caninumtrong thực hiện phản ứng PCR và giải trình tự gene trên đoạn gene ITS1. Xác định đa hình kiểu gene của loài sán dây Dipylidium caninumở các địa phương khác nhau qua hệ số di truyền và ma trận di truyền và so sánh trình tự DNA của sán dây Dipylidium caninumvới các mẫu đăng ký trên Genbank, vị trí của sán dây Dipylidium caninumở ĐBSCLso với GenBank trong cây phát sinh loài.

Mô tả vòng đời ký chủ trung gian sán dây D.caninum, mô tả vòng đời của loài sán dây Dipylidium caninum trên chó tại ĐBSCL và mô tả biến đổi bệnh lý học của sán dây Dipylidium caninum gây ra trên chó.

  1. Ý nghĩa thực tiễn và khả năng ứng dụng của luận án:

Là công trình nghiên cứu có hệ thống về sán dây trên chó: xác định tình hình nhiễm bệnh và các yếu tố liên quan đến sự phân bố của mầm bệnh. Định loài bằng hình thái học và sinh học phân tử sử dụng kỹ thuật PCR và giải trình tự gen ITS1.

Đã nghiên cứu thành công vòng đời Dipylidium caninum ở ĐBSCL, xác định thành phần ký chủ trung gian, nghiên cứu bệnh lý lâm sàng, điều trị thử nghiệm và đề ra biện pháp tẩy trừ.

Cung cấp thêm tư liệu khoa học về loài sán dây trên chó ở ĐBSCL, đồng thời bổ sung cho giáo trình ký sinh trùng thú y để phục vụ công tác giảng dạy.

Kết quả đề tài là cơ sở khoa học để khuyến cáo người dân ở vùng ĐBSCL áp dụng biện pháp phòng trị bệnh sán dây trên chó một cách hữu hiệu.

  1. Thesis summary

The thesis consists of five contents identifying the prevalence of tapeworm infection in dogs in the Mekong Delta and the life cycle of D. caninum from the oncosphere stage to the adult stage in the main host. This thesis also contributes to the description of clinical symptoms, pathology and histopathology and screening of anthelminthic treatment.

Research results

In 6 provinces of the Mekong Delta dogs were infected with tapeworm with the overall infection rate of 22,51% ((for fecal examination)) and 25.86% (for necropsy method), with 5 species which belong to 2 orders Cyclophyllidea and Pseudophylidea that were consisting of: Dipylidium caninum, Taenia pisiformis, Taenia hydatigena, Spirometra mansoni, Diphyllobothrium latum. Analysis of  Dipylidium caninum identification by morphological characteristics and molecular biology techniques through the amplification of  ITS and 28S gene regions. The study collated the sequence of nucleotides of the ITS gene segment from the same gene segment in the GenBank with 99% of the tapeworm genotypes in the study compared with the tapeworm samples registered in the GeneBank.

The results of the thesis determined that the time to complete the life cycle of the Dipylidium caninum tapeworm has 2 stages inside the intermediate host (Ctenocephalides) and inside the definitive host (dog).

Pathological research in dogs infected D. caninum with tapeworms shows common clinical symptoms such as itchy anus, diarrhea. The dog with D. caninum tapeworm diseases often appears to have the most common gross lesions in the small intestine which are localized hemorrhage at the site of the hook of the tapeworm, inflammatory bowel. Microscopic lesions in the digestive system of dogs infected with Dipylidium caninum tapeworm that showed lesions in the mucosal surface of the small intestine, intestinal muscle and mesenteric lymphadenitis. These typical injuries were peeling off villi of intestinal myositis, presence of many leukocytes in the submucosal of small intestine, fibrinous on mucosal surfaces, enlarged lymph nodes.

          Praziquantel and niclosamide can treat D. caninum dogs tapeworm disease with 100% cure rate at 10mg/kg for praziquantel and 150 mg/kg body weight for niclosamide.

  1. Contributions of the thesis

- The thesis has determined the epidemiological characteristics of tapeworm in dogs and identified some risk factors related to the rate of parasitic tapeworm infection, quantified the risk factors related to the rate tapeworm infection in dogs is equal to the odds ratio (Odd ratio).

- Identify the primers to identify Dipylidium caninum tapeworms in PCR and sequencing of ITS1 gene segments. Identify genotypic polymorphisms of Dipylidium caninum tapeworms in different localities through genetic coefficients and genetic matrix and compare DNA sequence of Dipylidium caninum tapeworm with GenBank registration forms of Dipylidium caninum tapeworm in Mekong Delta compared with GenBank in phylogenetic tree.

- The thesis has identified the intermediate host life cycle of Dipylidium caninum tapeworm, describes the life cycle of Dipylidium caninum in dogs in the Mekong Delta and describes the pathological changes of the Dipylidium caninum tapeworms dog.

  1. The practical significance and feasibility of the thesis: This is a systematic study on D. caninum tapeworm in a dog with regard to identifying the prevalence of infection and factor related to the pathogen through morphological and molecular characteristics (PCR and sequencing). The life cycle of D. caninum in dog in Mekong Delta was confirmed by identifying intermediate host (flea), clinical symptoms and anthelmintics testing for diagnosis and treatment. The findings of this research not only provide scientific reference about tapeworm in dogs in Mekong Delta but also supplement veterinary parasitology textbooks for teaching practices.  The findings of this study present the scientific evidence for recommending farmers' diagnosis, treatment and prevention from dogs tapeworm.
