  1. Tóm tắt nội dung luận án

          Đề tài: “Nghiên cứu pheromone giới tính và kairomone trong quản lý tổng hợp sâu tơ, Plutella xylostella Linnaeus (Lepidoptera: Plutellidae) hại rau cải” được thực hiện từ năm 2015 đến năm 2019 đạt được kết quả sau:

  • Tổng hợp pheromone giới tính của sâu tơ

Thông qua phản ứng Wittig chọn lọc làm phản ứng chính đã thành công trong việc tổng hợp hợp chất (Z)-11-hexadecen-1-ol (Z11-16:OH), tổng sản lượng 42,9% (từ 11-bromo-1-undecanol đến Z11-16:OH). Oxy hóa hợp chất Z11-16:OH trải qua phản ứng oxy hóa bằng PCC hoặc acetyl hóa bằng acetic anhydride thu được hợp chất Z11-16:Ald  và Z11-16:OAc (hiệu suất phản ứng 90,4 và 71,4%, tương ứng).

- Đánh giá hiệu quả hấp dẫn của pheromone tổng hợp đối với sâu tơ

+ Thí nghiệm ngoài đồng đánh giá ảnh hưởng của các thành phần pheromone giới tính tổng hợp đối với sâu tơ cho thấy rằng Z11-16:Ald và Z11-16:OAc là hai thành phần rất cần thiết cho sự hấp dẫn thành trùng sâu tơ, còn Z11-16:OH là thành phần bổ trợ giúp tăng cường hiệu quả hấp dẫn của mồi.

+ Ảnh hưởng của hàm lượng thành phần Z11-16:OH lên hiệu quả hấp dẫn của mồi pheromone giới tính tổng hợp đối với sâu tơ. Kết quả này cho thấy hợp chất Z11-16:OH với tỷ lệ 1-10% trong thành phần pheromone giới tính tổng hợp đã làm gia tăng đáng kể hiệu quả hấp dẫn đối với thành trùng sâu tơ, P.xylostella.

            + Ảnh hưởng của hàm lượng mồi lên hiệu quả hấp dẫn của mồi pheromone giới tính tổng hợp Z11-16:Ald, Z11-16:OAc và Z11-16:OH. Kết quả cho thấy rằng thành trùng đực sâu tơ cho đáp ứng mạnh với pheromone giới tính ở nồng độ thấp từ 0,01 đến 0,1 mg/tuýp, hiệu quả hấp dẫn bắt đầu giảm khi nồng độ cao hơn từ 0,3 đến 1,0 mg/tuýp.

            - Đánh giá hiệu quả hấp dẫn của kairomone đối với sâu tơ

            + Đánh giá hiệu quả của hợp chất Allyl isothiocyanate (AITC) đối với sâu tơ. Bẫy mồi với AITC nồng độ 0,5; 0,7 và 1,0 mg/tuýp cho hiệu quả hấp dẫn mạnh cả hai giới của thành trùng. Trong đó, nồng độ 0,7 mg/tuýp cho hiệu quả hấp dẫn cao hơn ý nghĩa so 1,0 ml dịch lá cải nghiền;

+ Đánh giá hiệu quả của hợp chất cis -3-hexenyl acetate (Z3-6:OAc) đối với sâu tơ. Bẫy với Z3-6:OAc nồng độ 0,01 và 1,0 mg/tuýp cũng cho hiệu quả hấp dẫn cao hai giới của thành trùng, tương đương với 1,0 ml dịch lá cải nghiền.

- Đánh giá hiệu quả hấp dẫn phối hợp của pheromone giới tính và kairomone đối với sâu tơ.

Kết quả sự kết hợp của pheromone giới tính tổng hợp và hợp chất Z3-6:OAc hoặc hợp chất AITC (tỷ lệ 1:1:70; 1:1:0; 1:0:70, tương ứng) hấp dẫn được cả hai giới của thành trùng đồng thời làm tăng sự hấp dẫn của thành trùng so với mồi chỉ gồm AITC và Z3-6:OAc riêng lẻ hoặc kết hợp.

- Ứng dụng pheromone giới tính tổng hợp và kairomone trong quản lý sâu tơ

+ Khảo sát thời gian hoạt động của thành trùng sâu tơ cho thấy rằng thành trùng sâu tơ hoạt động tăng mạnh từ 18:00 đến 19:00 giờ.

+ Khảo sát sự biến động mật số quần thể sâu tơ trong năm cho thấy thành trùng sâu tơ, P. xylostella hiện diện quanh năm tại 3 điểm khảo sát. Số lượng thành trùng sâu tơ vào bẫy cao điểm ở các thời điểm ngày 04/8/2027, từ ngày 02/02/2018 đến 16/03/2018, và ngày 16/03/2018. Ngược lại, mật số thành trùng hiện diện rất ít tại thời điểm từ 29/9/2017 đến 10/11/2017. Tương tự, mật số thành trùng vào bẫy cũng thấp ở tại 3 địa điểm khảo sát, thời điểm từ 30/03/2018 đến 11/5/2018.

+ Khảo sát sự biến động mật số quần thể và tỷ lệ rau cải bị hại do sâu tơ trong một vụ cải. Diễn biến mật số và sự gây hại của sâu tơ được ghi nhận trên các ruộng cải bắp, cải bông, cải bắp de và cải ngọt bằng cách đặt bẫy pheromone giới tính và khảo sát đồng ruộng. Sâu tơ xuất hiện và gây hại trong suốt vụ cải với diễn biến mật số thành trùng và tỷ lệ gây hại tương tự nhau, thấp ở giai đoạn đầu vụ và tăng dần cho đến trước khi thu hoạch (trừ cải bông). Hơn nữa, số lượng thành trùng đực vào bẫy tương quan chặt với tỷ lệ lá bị hại và tỷ lệ diện tích lá bị hại (hệ số tương quan r = 0,74 - 0,98).

+ Đánh giá hiệu quả của biện pháp đặt bẫy hấp dẫn (pheromone giới tính kết hợp AITC) đối với sâu tơ tại Tp. Đà Lạt, tỉnh Lâm Đồng và huyện Mỹ Xuyên, tỉnh Sóc Trăng. Kết quả biện pháp đặt bẫy kết hợp 2 lần phun thuốc sâu có hiệu quả trong quản lý sâu tơ ở điều kiện ngoài đồng.

  1. Những đóng góp mới của luận án

- Đề tài là công trình nghiên cứu đầu tiên về quy trình tổng hợp các thành phần của pheromone giới tính sâu tơ theo hướng ngắn gọn, điều kiện phản ứng đơn giản và dễ thực hiện, các hóa chất tổng hợp khá an toàn so với các quy trình trước đây.

- Lần đầu tiên ở ĐBSCL, sử dụng pheromone giới tính và kairomone để đánh giá hiệu quả khả năng hấp dẫn thành trùng sâu tơ ở điều kiện ngoài đồng.

- Bước đầu đánh giá hiệu quả kết hợp của pheromone giới tính và kairomone trong phòng trừ sâu tơ ở điều kiện ngoài đồng theo hướng an toàn nhằm góp phần giảm thiểu hoặc thay thế dần thuốc trừ sâu hóa học.

  1. Brief content of thesis

The thesis: “Research about sex pheromone and kairomone for integrated pest management of the diamondback moth, Plutella xylostella Linnaeus (Lepidoptera: Plutellidae) in vegetable”

- Synthesis of sex pheromone components of P. xylostella

The synthetic route using Wittig reaction as key reaction successfully synthesized (Z)-11-hexadecen-1-ol (Z11-16:OH) in 42.9% overall yield (from 11-bromon-1-undecanol to Z11-16:OH). Following Pyridinium chlorochromate (PCC) oxidation and acetylation of Z11-16:OH gave Z11-16:Ald and Z11-16:OAc (90,4% and 71,4% yield, respectively).

- Evaluation of the attractive effect of synthetic pheromones to P. xylostella

  + Field effects of synthetic sex pheromone composition to P. xylostella showed that Z11-16:Ald and Z11-16:OAc are essential for the attraction while Z11-16:OH was an auxiliary component for the attraction of P. xylostella males.

+ Effect of the content of Z11-16:OH on the attractive effect of synthetic sex pheromone to P. xylostella. This result shows that the compound Z11-16:OH with the ratio of 1-10% in the synthetic sex pheromone composition has increased significantly the attractive effect of P.xylostella males.

  + Effect of concentration on attractive effect of synthetic sex pheromone lure Z11-16:Ald, Z11-16:OAc and Z11-16:OH for P.xylostella. This result shows that the P. xylostella males gave a strong response to sex pheromone lure at low concentrations from 0.01 to 0.1 mg/type, then the attractive effect starts to decrease gradually as the concentration gets higher from 0.3 to 1.0 mg/tube.

  - Evaluation of the attractive effect of kairomone to P. xylostella

  + Evaluation of the attractive effect of Allyl isothiocyanate (AITC) to P. xylostella. The traps baited with AITC (0,5; 0,7 and 1,0 mg/lure) showed that high attraction of the moths of both males and females.  In which, the bait with the concentration of 0.7 mg/tube (D-4) had the strongest effect on attracting the diamondback moth, significantly different from the lure with 1.0 ml of leaf grinded solution.

+ Evaluation of the attractive effect of cis -3-hexenyl acetate (Z3-6:OAc) to P. xylostella. Traps baited with Z3-6:OAc (0,01 and 1,0 mg/lure) showed high attraction of the moths of both sexes and as strong as the a brassica leaf grinded solution did in our field tests.

- Evaluation of the combined attractive effects of sex pheromone and kairomone to P. xylostella. This result demonstrates that the combination of the synthetic sex pheromone and the Z3-6:OAc compound or the AITC compound (1:1:70; 1:1:0; 1:0:70, respectively) is highly attractive both P. xylostella males and female while increasing the attractiveness of adult compared with lures consisting of AITC and Z3-6:OAc single or in combination.

- Application of synthetic sex pheromone and kairomone in management to P. xylostella

+ Investigate the activity time of P. xylostella shows that males activity increased sharply from 18:00 to 19:00 hours.

+ Survey on changes in population density of P. xylostella during the year. The number of P.xylostella entering the trap peaked at the time of 4th August 2027, from 2th February2018 to16th March 2018, and on 16th March 2018. In contrast, the density of P.xylostella was very low from 29th September 2017 to 10th November 2017. Similarly, the density of larvae entering the trap was also low at the 3 survey sites, from 30th March 2018 to 11th May 2018.

+ Survey on population density variation and percentage of vegetables damaged by P. xylostella in a planting crop

Dynamics of population and damage of P. xylostella were monitored at the cabbage, cauliflower, De cabbage and leaf mustard field . fields by use of sex pheromone traps and field survey. P. xylostella presented and damaged at surveyed fields through out the crop with the dynamics of population and damage kept at low level at the early crop season and then gradually increased until before harvest (exception at broccoli field). Furthermore, the numbers of captured males tightly correlated with damage ratios (r = 0.74- 0.98).

In the file, for the efficacy of traps baited with a synthetic mixture of sex pheromone (5:5:1; 0,01 mg/lure) and AITC (0,7 mg/lure)  has been effective for the control of P. xylostella in brassicaceae.

  1. Contribution of thesis

- It is the first research to synthetic sex pheromones of the diamondback moth, P. xylostella by short, simple reaction conditions, easy to perform and safe synthetic chemicals.

- For the first time in the Mekong Delta, sex pheromone and kairomone were used to evaluate the effectiveness of attracting the diamondback moth in field.

- Initial evaluation of the combined effectiveness of sex pheromone and kairomone in controlling the diamondback moth in field conditions in a safe direction to contribute to reducing or gradually replacing chemical pesticides.
