The development of vaccines for striped catfish (Pangasianodon hypophthalmus) in the Mekong Delta is an urgent need due to the diversity of bacterial pathogens and the increasing prevalence of antimicrobial resistance. This study aimed to evaluate the feasibility and efficacy of injectable autogenous vaccines for the prevention of hemorrhagic septicemia and bacillary necrosis of internal organs in striped catfish.
A total of eight Aeromonas spp. strains, including three A. hydrophila and five A. dhakensis, and eleven Edwardsiella ictaluri strains were identified from diseased fish using 16S rRNA gene sequencing. DNA fingerprinting and virulence gene analysis revealed clear differentiation between Aeromonas species, whereas E. ictaluri exhibited a relatively conserved virulence gene profile with some degree of variability.
Principal component analysis indicated that bacterial differentiation was primarily driven by genetic factors. Aeromonas isolates were grouped into two major clusters corresponding to A. hydrophila and A. dhakensis, while E. ictaluri showed higher similarity among strains.
Based on epidemiological characteristics, virulence, and genetic diversity, representative strains were selected for autogenous vaccine development, including A. hydrophila VH31, A. dhakensis VH74, E. ictaluri VH83, and E. ictaluri VH116. A synergistic effect in virulence was also observed during co-infection experiments.
Challenge trials demonstrated that multivalent vaccines provided high protective efficacy (87–100%), significantly outstanding monovalent vaccines. In addition, the vaccines induced both specific and non-specific immune responses, maintained stable hematological parameters, and exhibited cross-protective immunity among bacterial strains.
These findings highlight the potential of multivalent autogenous vaccines in controlling hemorrhagic septicemia and bacillary necrosis in striped catfish, emphasizing the critical role of strain selection and combination in optimizing vaccine efficacy. Further studies under field conditions are recommended to validate and refine the application process.
The dissertation elucidated the differentiation between Aeromonas and Edwardsiella bacterial pathogens in striped catfish affected by hemorrhagic disease and bacillary necrosis of the liver and kidney through an analysis of overall differences, thereby providing a novel approach to the study of bacterial pathology in fish.
Notably, the dissertation identified synergistic virulence during dual infection with Aeromonas strains and Edwardsiella ictaluri, providing scientific evidence that disease pathogenesis in striped catfish involves multiple pathogens rather than a single causative agent.
Furthermore, the dissertation demonstrated that incorporating bacterial strains directly associated with the disease pathology into the development of autogenous vaccines can improve protective efficacy. The study also documented cross-protection among bacterial strains, contributing to a better understanding of the scientific basis for optimizing vaccine composition in aquaculture disease prevention.
The findings of this dissertation provide a practical basis for improving the diagnosis and differentiation of pathogens in striped catfish, thereby supporting the selection of appropriate disease prevention and treatment strategies under actual aquaculture conditions.
The proposed integration of bacterial screening methods facilitates the selection of suitable antigens, laying the groundwork for the development of highly effective autogenous vaccines tailored to the epidemiological conditions of individual farming regions.
Furthermore, the research findings offer potential applications in the design of vaccines based on local disease characteristics, contributing to improved disease prevention, reduced mortality, and decreased antibiotic use in aquaculture.
Overall, this dissertation has practical significance in supporting farmers and hatcheries in effective disease control, minimizing economic losses, and promoting the sustainable development of the striped catfish farming industry.
